Kalou has been absent with a thigh injury
Chelsea are still without striker Didier Drogba who has failed to recover from a virus while defender Alex and midfielder Frank Lampard also miss out.
The Blues welcome back forwards Daniel Sturridge and Salomon Kalou for Tuesday's game on the artificial pitch.
Spartak are without key midfielder Alex due to a thigh injury but Brazilian striker Welliton is expected to recover from a similar injury.
Republic of Ireland winger Aiden McGeady is also set to start in Moscow.
Chelsea will be looking to banish the nightmare of their last visit to the 80,000-seater Luzhniki Stadium, where they lost the 2008 Champions League final to Manchester United on penalties.

Captain John Terry's missed spot-kick paved the way for a United victory after the game had ended 1-1 following extra time.
Terry slipped on the wet grass as he went to take his penalty but the surface in the stadium pitch is now artificial.
The artificial pitch has led manager Carlo Ancelotti to delay picking his team and will instead see how his side train on the surface in Moscow.
"I can't give the line-up because we want to try the pitch before," said Ancelotti.
"If the players are comfortable to play there I will take the decision after the training session."
Meanwhile Chelsea left-back Ashley Cole has lost two Champions League finals, one with Arsenal in 2006 and another with Chelsea in the Luzhniki two years ago.
"To this day I still think about the Champions League finals that I've lost," said Cole.
"Little things like that come into your head, and it makes you more determined to go out and train harder with the guys every day."
Meanwhile, the two victories for the Muscovites in Group F has given their fans renewed belief that they can qualify for the knockout stage.
"We have had a great start, probably better than some of our fans had expected," said manager Valery Karpin.
"But we all know that beating Chelsea would be a totally different proposition from what we had before. So we're not getting carried away by any means.
"I don't want to predict the result. We have made a good start and are well positioned in the group now. We will need to put the handcuffs on [Didier] Drogba and [Nicolas] Anelka."
